* [[Geraldine Page]] kama Madame Medusa, a greedy, redheaded, wicked [[pawnshop]] owner and the lead villainess in ''The Rescuers''. Upon discovering the Devil's Eye diamond hidden in a [[blowhole (geology)|blowhole]], she kidnaps the small orphan, Penny, to retrieve it for her. In the end, she is thwarted and presumably eaten by her two [[alligators]], Brutus and Nero. Originally, [[Cruella de Vil]] from ''[[101 Dalmatians (1961 film)|101 Dalmatians]]'' was to have been recast as the villainess in ''The Rescuers'', but this idea was dropped since the studio was not interested in producing sequels at the time, and thus Cruella was replaced by a retouched version of the Diamond Duchess in ''Miss Bianca''. The two characters share surprisingly few similarities, other than perhaps the tendency to drive recklessly.

* [[Michelle Stacy]] kama Penny, a lonely [[orphan]] girl, residing at Morningside Orphanage in [[New York City]]. She is comforted by her [[cat]], Rufus, who tells her to have faith, and is kidnapped by Medusa in an attempt to retrieve the world's largest diamond, the Devil's Eye. She was likely based off of Patience, the orphan in Margery Sharp's ''Miss Bianca'' novel.

* [[Jim Jordan]] kama Orville, an [[albatrosi]] who gives Bernard and Bianca a ride to Devil's Bayou. Originally, a pigeon had been proposed as the mice's transportation, until animator [[Frank Thomas (animator)|Frank Thomas]] suggested an albatross due to its lumbering takeoff. The role was the last for Jim Jordan, who retired after the film's release and died later in 1988.

* [[John McIntire]] kama Rufus, Rufus is the elderly cat who resides at Morningside Orphanage, often tending to Penny, when she is sad. Although his time onscreen is rather brief, he provides the film's most important theme, [[faith]]. He was designed by animator [[Ollie Johnston]], with this being his last animation role after employing for Disney for 40 years.
